Amyloidaggregationofα-synuclein(AS)hasbeen linked to the pathological effects associated with IIParkinson?sdisease(PD).CubindsspecificallyattheN-IterminusofASandtriggersitsaggregation.Site-specificCu-catalyzedoxidationofAShasbeenproposedasaplausiblemechanismformetal-enhancedASamyloidformation.InthisIstudy,CubindingtoASwasprobedbyNMRspectroscopy,incombinationwithsyntheticpeptidemodels,site-directedmutagenesis,andC-terminal-truncatedproteinvariants.OurresultsdemonstratethatbothMetresiduesinthemotif15MDVFMconstitutekeystructuraldeterminantsforthehigh-IaffinitybindingofCutotheN-terminalregionofAS.
