Dendritic cells can function as adjuvants able to mediate protection against differentpathogens. Given that successful vaccination against F. hepatica is mostly related to theinduction of Th1 responses, we studied the potential of DC loaded with F. hepatica Agsand LPS (which promote DC maturation) as a vaccine against fasciolosis in BALB/cmice. However, only a semi-mature phenotype was achieved when DC weresimultaneously cultured with a F. hepatica total extract (TE) and LPS. The activationstatus of TE loaded DC was enhanced when these cells were treated with TE 90 minbefore being stimulated with LPS (TE90 DC). More importantly, a single immunizationof mice with TE90 DC stimulated a systemic Th1 response and conferred protectionagainst hepatic damage induced by F. hepatica infection. Thus, TE90 DC may prove tobe a useful new tool for vaccination against F. hepatica.
