Resumen:
Senecio genus is spread worldwide, being some species poisonous due tothe presence of pyrrolizidine alkaloids (PA). Poisoning in humans is associatedto the consumption of herbal teas, while in domestic animals to theintake of plants during dry season. PA are alkylating agents with antimitoticeffect, causing liver megalocytosis, ductal proliferation and fibrosis. InArgentina, Senecio grisebachii Baker (?margarita de campo?, SG) grows inCentral and Northeastern regions. Aim of this work is to present effects offlower and leaf extracts of SG on cell viability. Extracts were prepared with15 g of dried flowers or leaves and 200mL methanol using magnetic stirrerfor 24 h. They were rotaevaporated until dryness and re-suspended with1% HCl, alkalizing with 25% NH3. Further extraction included chloroform:methanol (4:1) and evaporation to dryness under N2, re-suspendingwith 10 mL DMSO. Presence of PAwas performed with Ehrlich reagent andTLC. MTT assay was used to assess cell viability, using bovine lymphocytesstimulated with 100 mg/mL phytohemagglutinin. Cells were cultured inHam F12 supplemented with fetal bovine serum and antibiotics at 37 Cduring 48 h. Treatments were performed on the last 24 h. Absorbancedetermination was performed with Elisa at 550 nm. Treatments were: 1)negative control 2) positive control (methanol 10%) 3) diluent control (0,1%DMSO); and S. grisebachii extracts 4) 25 mg/mL 5) 50 mg/mL 6) 100 mg/mLand 7) 200 mg/mL.
