Nicotiana glauca Graham (Solanaceae) ?Palán - Palán?, is used popularly in topical treatments However, Anabasine is the main alkaloid in this specie, which has caused some fatal cases by accidental consumption. The aim of this work is to develop a rapid and sensitive Flow injection analysis (FIA)-spectrofluorimetric methodology for Anabasine quantification in herbal medicines, as adulterant. The sample solution consisted in an extract from commercial herbal medicines (Peumus boldus M. ?Boldo?, Malva sylvestris L. ?Malva?) and an extract from Nicotiana glauca leaves was used like positive control. In order to reduce the potential matrix interferences and to enhance the method sensitivity, a pre-concentration system composed by a mini column filled with XAD-4 resin, was employed.The sample solution and the eluent were pumped through a FIA manifold with fluorimetric detection (λex = 314 nm and λem = 440 nm, slit width ex/em: 5/5 nm) using a 120 mL flow cell unit. The studied experimental variables that influenced on FIA system were sample pH, sample flow rate, and eluent flow rate. These variables were analyzed by Design Expert 6.0.8 program in order to optimize the experimental conditions.The obtained analytical parameters were: regression equation y = 360.68x + 1.6019 with were R2= 0.9922; limits of detection and quantification were 4.22 x 10 -6 mg L-1 and 1.28 x 10 -5 mg L-1. In all the determinations, the standard deviation was better than 5.9 x 10 -2. The found Anabasine concentration in the positive control was 4.74 x 10-3 mg L-1, while in the samples the Anabasine concentration was negative.The proposed simple methodology was applied to the determination of the alkaloid in methanolic extracts from Nicotiana glauca leaves. The sensitivity of the methodology was adequate for the quantification of analyte traces.
