Resumen:
Serum bile acids´ (SBA) are sensitive markers of cholestasis, however, reference chromatographic methods used in the analysis of individual bile acids (GC and HPLC) are expensive, complex and require sequential steps. Capillary electrophoresis is a versatile and highly selective microanalysis technique. Patients and methods: 23 patients with chronic renal failure, 15 renal transplant recipients, 20 patients with intrahepatic cholestasis of pregnancy, 5 with hyperthiroidism associated cholestasis and 27 healthy adults (control s) were, studied. Free _nd conjugated forms (glycine or taurine) of ursodeoxicholic, cholic, lithocholic, deoxycholic and chenodeoxycholic acids were measured in serum samples by capillary ion analyzer (Waters, Milford MA, USA). Micellar electrokinetic chromatography was performed with sodium dodecyl sulphate 50 mM,pH 7 modified with sulphatade and hidroxypropyl cyclodextrines 5 mM, 25 kV, at 30°C and 185 nm for detection using a capillary of 60 cm length and}5 11m I.D. The complete procedure was carried out ror each sample in 50 minutes. AlI patients showed elevated levels of SBA, with characteristic profiles. In conclusion, a very simple and fast capillary electrophoretic method, suitable ror . the simultaneous determination of 15 bile acids (in their free, glyco- and tauro- derivative forms) with good precision and accuracy may be useful in the diagnosis of colestatic . diseases.
