EVENTS ASSOCIATED TO ORAL ADMINISTRATION OF OLIGODEOXYNUCLEOTIDES CONTAINING CpG MOTIFS (CpG)

CpG is an effective adjuvant through different mucosal routes. However, the early effects, responsible for the final outcome of the immune response, have not been studied. We proved that CpG is an effective adjuvant to induce local and systemic OVA-specific immune response after oral administration. Regarding the changes in gut-associated lymphoid tissues and spleen from Balb/c mice, we observed that 24h after oral administration of CpG there was a higher cellularity in Peyer’s patches (PP) but not changes in mesenteric lymph nodes (MLN) or spleen compared to saline-treated mice. Furthermore, only a slightly increased percentage of CD19+ cells in PP (saline: 63%; CpG: 68%) and MLN (saline: 16%; CpG: 21%) were observed by flow cytometry. Moreover, an increase in the expression of CD86 (MFI: saline: 13; CpG: 21) and MHC II (MFI: saline: 471; CpG: 650) on CD19+ cells was found in MLN of CpG-treated mice. In addition, cells from CpG-treated mice produce higher levels of IL-12 (pg/ml) in culture supernatants of PP (saline: 1199; CpG: 2285), MLN (saline: 1052; CpG: 1407) and spleen (saline: 3858; CpG: 4715) after in vitro CpG stimulation.
