MISTIC2: Meta-server to calculate covariation and visualize MSA information.

MISTIC2 is an upgrade to the Mutual Information Server to Infer Coevolution: It is a complete tool to calculate covariation, analyze Multiple Sequence Alignments information and explore results in a rich graphical interface. MISTIC2 not only calculates corrected MI (MIp) but has been upgraded to incorporate new Direct Coupling methods such as mfDCA, plmDCA and gaussianDCA. At the same time it remains flexible to include new algorithms that may be developed in the near future.The server provides two ways of accession, an Application Programmable Interface (API) and a Graphical User Interface. The programmable interface returns raw results, while the graphical interface presents results visualization and several information-related quantities that can be interactively explored. MISTIC2 provides an integrated view of the MSA in terms of (i) the covariation scores between residue pairs (MI, mfDCA, plmDCA and gaussianDCA), (ii) sequence conservation and (iii) residue-level cumulative and proximity covariation scores. It allows the immediate inter-comparison of all methods in the same framework and at the same time it can integrate multiple covariation results into a combined score. Several tools are offered for selecting subsets of nodes (residues) from the network that can be automatically visualized in a representative 3D structure, while coloring can be set to match different attributes.Finally, the raw results can be downloaded to further analysis from the visualization interface. The server is available at http://mistic2.leloir.org.ar. In summary, MISTIC2 allows for a comprehensive, compact, visually rich view of the information contained within an MSA unique to any other publicly available web server.
