Selection improvement by ligand excess (SIMPLEX) was developed to select a specific aptamers for a selected target from a combinatorial library in only one partition step. The aim of this procedure is to obtain more specific than non specific ligands in the selection.  This present work is a new in vitro method, where the enzymatic amplification was applied to the library before the selection step (Scheme 1). It is shown how this treatment produces the desire effect on the selection step, increasing the probability to obtain DNA aptamers able to recognize its target. The affinity and specificity of the aptamers obtained were proved by filter binding assays and used for protein detection in histologies or purification from complex mixtures coming form mice brains.
