Resumen:
More than 1% of human genes contribute to cancer that arisesdue to somatic mutations. These confers tumor cells a growthadvantage and are known as ?driver? mutations. Activatingmutations in proto-oncogenic Protein Kinases (PKs) arefrequent driver events in many human tumor types. Drivermutations often occur at frequencies indistinguishable fromthose of passenger mutations and discriminating them is asignificant challenge facing the fields of genomics, cancerbiology and therapeutics. Furthermore, there are no curatedsets of driver and passenger cancer alterations that could beused to test methods. Recent findings in our group (manuscriptin press) demonstrate that current methods and tools toidentify driver mutations (as MutationAssesor (1), transFIC (2)and FATHMM (3)) based on conserved residues, systematicallyfail to discriminate most activating mutations of protooncogenicPKs.
