Resumen:
We introduce here a new fluorescent derivative of 1-thio-β-N-acetylglucosamine linked to a pyrene systemthroughatriazolylpentylspacer,designedtoself-assembleintoamultivalentglycocluster.ThesynthesiswasachievedbyefficientCuAACclickreactionbetweenapyrenefunctionalizedwithanazidegroupandasuitablealkynylthiomonosaccharide.Spectroscopicstudiesbyfluorometryindicatedthattheself-assemblyinaqueousmediumismodulatedbyconcentrationandpHchanges,thelatterduetothepresenceoftheaminogroupclosetotheπsystem.Circulardichroismexperimentsrevealedamoderatepositivesignal,suggestingthatthepyrene-thioGlcNAcconjugatecanaggregateintoachiralsupramolecularassembly.Thesugarmoietyshowedtospe-cificallyandreversiblyinteractwiththewheatgermagglutinin,afactthatwasdemonstratedbyturbidityassay.SEMmicroscopyofalyophilizedsolutionatpH10revealedafibrillarmorphologycompatiblewiththepresenceoftubularmicelles,whereascrystallineandamorphoussolidsareformedatlowerpHs
