PRODUCTION OF THE 14D9 ANTIBODY BY NICOTIANA TABACUM CELL SUSPENSION BATCH CULTURE IN A 2L-BIOREACTOR

Resumen:
We have performed the
scale up of a Nicotiana tabacum cell
suspension culture of the transgenic line Ab-KDEL expressing the
recombinant catalytic antibody 14D9 (r14D9) from 225 ml-Erlenmeyer flasks to a
2L-bioreactor. To set up the batch processes, volumetric oxygen transfer
coefficient (KLa) was adjusted to 42.7/h both in Erlenmeyer flasks and
bioreactor. The influence of different inoculum sizes (1%, 5% and 10% w/v) on
growth and antibody production was tested. The maximal specific growth rate
(0.150/d), doubling time (5d), biomass (78.5g/l), maximal r14D9 productivity
(1.31 µg
r14D9/g DW.d), antibody accumulation in the biomass
(157 µg r14D9/mg) and in the culture medium ( 8.7 µg r14D9/mL) were achieved at
1% (w/p) inoculum size
