MOLECULAR CHARACTERIZATION OF THE GENUS FRAGARIA INTERSPECIFIC HYBRIDS

In the Active Germplasm Bank (BGA) of strawberry (Tucumán, Argentina), were performed out crosses between wild species of the genus Fragaria, F. chiloensis x F. virginiana. To obtain fertile intraspecific hybrids, better adapted to environmental region conditions, the reproductive behavior of the genotypes present in the BGA is analyzed. Using the microsatellites technique, the molecular characterization is performed in four putative hybrid genotypes obtained in crosses. DNA was extracted from frozen at -70 ° C young leaves and grinded with liquid nitrogen, using for the extraction the Plant Genomic DNA Miniprep GenEluteTM Kit. The samples were amplified with primers SSR, and the amplification conditions were adjusted. The amplified products were separated by electrophoresis in agarose gel and visualized with ethidium bromide. The genotypes analyzed FCFVa; FCFVb and FCFVd showed bands of the male parent, using only three pairs of primers. In the case of FCFVc genotype, we used 15 pairs of primers, showing only the female parent bands. These results suggest that genotypes FCFVa, FCFVb and FCFVd would be hybrids of crosses between F. chiloensis x F. virginiana. In the case of FCFVc genotype, could be an individual produced by apomixis, a common phenomenon in this group of plants.
