Enzymatic hydrolysis of naringin for the production of prunin and rhamnose was studied under alkaline conditions. A partially purified α-rhamnosidase (Rhase) from Verticillium tenerum free of β-glucosidase (by heat inactivation) was used. Effects of pH and temperature on enzyme stability and activity were determined. Hydrolysis of naringin was carried out using 2 g/L substrate solutions, 0.8 mU of Rhase in 20 mM Tris-HCl buffer, pH 9.0 at 45ºC. Naringin, prunin and naringenin were determined by HPLC. Rhase is stable up to 48.5ºC and from pH: 6.0 to 11. Rhase activity presents a maximum at pH: 6.0, with a 37% of this activity at pH: 9.5. Under these reaction conditions, a 12.6 % naringin hydrolysis was obtained in 20 h, without naringenin production. These results show that Rhase from V. tenerum is a promissory alternative to produce rhamnose and prunin from naringin.
