In the dawn of the twentieth century Paul Ehrlich introduces the concept of magic bullet (magische Kugel), as an ideal chemotherapeutic agent that would selectively target a disease-causing organisms without impairing its host. One century later, the challenge to bring ?magic bullets? is still in the mind of scientists. The concept of targeted therapeutics is intrinsically associated to the ideas of specificity and selectivity. In the drug design arena, the target-based paradigm pursues exquisitely selective drugs (e.g., drugs that present higher affinity for a given isoform of the target which is preferably expressed in the target organ, or anti-infective agents that can bind their targets in the diseasecausingorganism but not similar targets in the host).
