Respiratory diseases are a major health issue around the world. Local delivery of drugs to the lung tissue is complicated due to the unique architecture of the respiratory system and its natural defense mechanisms. New therapeutic strategies are needed in order to overcome these issues. Liposomal delivery systems that encapsulate mucolytics are an alternative; these substances break down the gel structure of mucus, helping to overcome barriers and increasing the effectiveness of different therapies. Based on this, the main goal of this work was to improve effectiveness of stem cell therapy in chronic lung diseases. Mucolytic transporters composed of lung surfactant phospholipids were developed to improve their interaction with the lung tissue. Liposomes were obtained by the dehydration/rehydration method. Extrusion was applied to obtain smaller vesicles, which were combined with mucolytics. Microscopy and dynamic light scattering described large unilamellar vesicles with a hydrodynamic ratio of 100 nm. Finally, lung mechanics were improved after intratracheal instillation of the formulations and bone marrow derived cells intravenous injection in a mice model of silicosis.
