Resumen:
BPA, an endocrine disruptor used in the manufacture of plastics andepoxy resins, could modulate the inflammatory immune response. Itis known that BPA may interfere with dopaminergic transmission atCNS but there are less evidences at peripheral level. We previouslydemonstrated that the treatment with BPA for 24 h did not modifythe production of IL-6 and IL-8 or the response to dopamine in humankeratinocytes and macrophages. In this work we evaluate theeffect of BPA, at long times, alone or in combination with dopamine(DA) on the cytokine production (IL-6, IL-8, IL-1β, ELISA), the NFκBpathway (Western blot) and the metalloproteinase (MMP) activity(Zymography) in a cell line of human keratinocytes HaCaT and THP-1 macrophages (PMA-differentiated cells). The cells were culturedwith BPA (10-7 M) for 10 days and then stimulated with DA (10-5and 10-4 M) for 24 h. In HaCaT keratinocytes, during the treatmentwith BPA no changes were observed in the levels of IL-6 nor in theactivation of NFκB induced by dopamine in the absence of the disruptor.However, the presence of BPA alters the effect of DA on theproduction of IL-8 and the MMP-9 activity (p
