arPlantcellvacuolesoccupyupto90%ofthecellvolumeandareconstantlysubjectedtowaterandsoluteexchange.Theosmoticflowandvacuolevolumedynamicsreliesonthevacuolemembrane-thetonoplast-anditscapacitytoregulateitspermeabilitytobothwaterandsolutes.Theosmoticpermeabilitycoefficient(Pf)istheparameterthatbettercharacterizesthewatertransportwhensubmittedtoanosmoticgradient.Usually,Pfdeterminationsaremadeinvitrofromtheinitialrateofvolumechange,whenafast(almostinstantaneous)osmolalitychangeoccurs.Whenaquaporinsarepresent,itisacceptedthatinitialvolumechangesareonlyduetowatermovements.However,inlivingcellsosmoticchangesarenotnecessarilyabruptbutgraduallyimposed.Undertheseconditions,waterfluxmightnotbetheonlyrelevantdrivingforceshapingthevacuolevolumeresponse.Inthisstudy,wequantitativelyinvestigatedvolumedynamicsofisolatedBetavulgarisrootvacuolesunderprogressivelyappliedosmoticgradientsatdifferentpH,aconditionthatmodifiesthetonoplastPf.Wefollowedthevacuolevolumechangeswhilesimultaneouslydeterminingtheexternalosmolalitytime-coursesandanalyzingthesedatawithmathematicalmodelling.
