A Ser678: Pro Substitution in Fks1p Confers Resistance to Echinocandin Drugs in Aspergillus fumigatus.

A point mutation was introduced in the Aspergillus fumigatus FKS1 gene. This gene is the target of echinocandin drugs. The amino acid change S678P in Fks1p confers a pattern of echinocandin cross-resistance. This phenotype was evaluated by in vitro susceptibility testing and also using Fks1p purified enzyme. This work demonstrate that Fks1p mutations are responsible of echinocandin resistance in Aspergillus fumigatus as in Candida spp.
