Murine perigestational alcohol ingestion
until day 10 of gestation (GD) increases resorption at DG7-8. We postulate a
link with MMP-2 and -9 expression changes and cellular alterations on GD8
implantation sites (IS). Ethanol/water-10% was administered to CF-1 females 17
days before and up to GD8 (TF) (Control Females, water). Differentiation stage,
histopathology, apoptotic index, collagen-IV and MMP-2 and -9 immunoexpression
were evaluated in decidua (De), ectoplacental cone (EC), trophoblast-decidual interface
(TDInt), embryo and extraembryonic tissues. TF showed 24h of differentiation delay.
Non-delayed-TF-De had decreased collagen-IV, altered MMP-2 and -9 expression and
increased TUNEL+cells. TF-EC-Proliferative area was lower and MMP-9 increased. TF-TDIn
had histological abnormalities and increased erythrocyte infiltration, apoptosis
and MMP-9 immunoexpression. TF-embryos (66%) had abnormal neuroectoderm with high
MMP-2 and -9. Periconceptional alcohol exposure produces histological defects, high
cell death and collagen-IV, MMP-2 and -9 expression changes at GD8, suggesting that
these factors participate on early embryonic loss induction.
