Preliminary results of the influence of the oocyte maturation media in the chromosomal numeric abnormalities on the in vitro derived early cattle embryos.

The aim of this study was to evaluate the influence of the oocyte culture media on the rate of numerical chromosome abnormalities in early bovine embryos derived from in vitro fertilization (IVF). Four different culture media (TCM-199; DME; H-Y and RPMI 1640), supplemented with BBS, FSH, LH estradiol and antibiotics were used in ths experiment. Groups of 150 COC?s were matured in each different media for 20hs at 38.5 °C in an atmosphere of 5 % CO2 in humidified air. After maturation, oocytes were in vitro fertilized for 18 h on IVF TALP media using a final sperm concentration of 1×106cells/ml. Presumptive zygotes were cultured for 72 h in SOF media. Early embryos were collected and fixed individually for chromosomal analysis. A total of 240 embryos were suitable for karyotiping. There were no statistically differences (p>0.05) in chromosomal alteration rates among embryos derived from oocyte cultured in different maturation media. Therefore, in our experiment, the oocyte culture media apparently does not affect the chromosomal abnormality rates of the derived embryos. However, there are previous studies that report different results in other species. Further experiments are currently in development to obtain more conclusive evidences in this issue.
