Resumen:
Administration of the erythroid survival factor erythropoietin (Epo) has been linked to the downregulation of hepcidin (Hep), a small peptide expressed mainly in the liver which impairs Fe release through the ferroportin exporter. We previously demonstrated that Epo (20 U/mL, 6 h) decreases Hep mRNA expression in the human liver HepG2 cell line, depending on the activation of the Epo receptor followed by the JAK2/PI3K/AKT/mTOR pathway. Hereby we aimed to determine which transcription factors downstream of this kinase cascade may play a role in Epo-induced Hep suppression. The transcription factor C/EBP-α is known to enhance Hep expression after a variety of stimuli. In HepG2 cells, Epo decreased C/EBP-α binding to the Hep promoter, as observed in electrophoretic mobility shift essays (EMSA, band densitometry: C=1, *E3h=0.7土0.1, n=4, p
