Radiotherapy (RT) is widely used in tumor treatment, but tumor cells sometimes acquire resistance resulting in recurrence and failure in many patients. The transcription factor NF-kB activates survival pathways in response to RT, proposing it as a therapeutic target to improve RT effect. Natural compounds of the flavonoid family have the ability to block NF-kB pathway. Therefore, we analyzed Silybin (Sb) flavonoid capability to enhance antitumor effect of RT on MB49 murine cells in vitro and in vivo. In vitro, RT (LD50 at 120h post-RT: over than 8Gy) and Silybin (LD50: 120μM) induced dose-dependent cytotoxicity; Silybin magnified RT inhibitory effect on cell viability (Viability. RT: 89%, Sb: 64%, RT+Sb: 31%, p=0.01) and clonogenic capacity (2Gy vs. 2Gy+Sb80μM p
