In a previous study we have demonstrated that Minthostachys verticillata essential oil (EO) modulate the pro-inflammatory cytokines expression in a mouse mastitis model at 72 and 96 h post-inoculation (p.i). The aim of this work was to determine the effect of EO on TLR2, TNF-α and IL-1β expression in a mouse mastitis model challenged with Enterococcus faecium at 24 and 48 h p.i. In addition, antibody production was evaluated at 360 h. Female Balb/c lactating mice were inoculated in mammary glands (MG) as follows: Group 1 (control): 100 µl of PBS/DMSO; Group 2: 100 μl of Enterococcus faecium (EF) (1x10 8 CFU/ml); Group 3: pretreatment with EO and challenge with EF. Gene expression levels in the MG by quantitative Real Time-PCR and EF-specific antibodies (IgM, IgG, IgA) in serum and MG homogenates by indirect ELISA, were evaluated. TLR2 and TNF-α expression were not detected in treated groups at the times assayed. No IL-1β expression was detected in group 2 at 24 h p.i.
