Although the molecular componentsand mechanical events driving later steps in this process are well understood,little is known about the molecular and cell processes that underlie retractionand cell rounding. We identify Moesin (the only member of theERM family in Drosophila) as a key player. We find that Moesin, whichcrosslinks actin filaments to the plasma membrane, is activated at the onset ofmitosis and is essential for cortical stiffening, retraction of the cell marginand rounding. Additionally, Moesin activation is sufficient to cause cellrounding and an increase of cortical stiffness in flat interphase cells.Finally, RNAi induced Moesin silencing reduction leads to aberrant spindlemorphology and abnormal chromosomes segregation, with a concomitant delay inthe onset of anaphase. We propose that Moesin plays a key role inmitotic progression by controlling cortical stiffness, cell rounding andmitotic spindle morphogeneis.
