Resumen:
Myelofibrosis (MF) and Essential Thrombocythemia (ET) are characterized by abnormal proliferation of hematopoietic progenitors and chronic inflammation. MF presents medullary fibrosis, which confers a worse prognosis to MF patients. Our objective was to evaluate the effect of plasma from MF and ET patients on megakaryo/thrombopoiesis.Megakaryopoiesis was evaluated in CD34+ hematopoietic progenitors (umbilical cord blood) cultured with TPO and 10% recalcified plasma (patients n=51, 29 MF and 22 ET; controls n=45) for 12 days. The number of cells and CD61 and CD42b percentage was quantified by Neubauer chamber and by flow cytometry, respectively. Thrombopoiesis was evaluated in mature megakaryocytes (MKs) cultured for 72 hours with TPO and 10% recalcified plasma. Proplatelets (PP) were counted by inverted microscope and its morphology was analyzed by immunofluorescence. Mann Whitney test and Spearman's correlation were used. An increase of MKs and their degree of maturity was observed in the presence of MF patients plasma compared to ET and control (p
