The Mal de Río Cuarto virus, one of the main diseases that affects maize crops in Argentina, is a member of the genusFijivirus (Reoviridae) whose genome is separated in 10 dsRNA segments. The genomic segments are arranged in a characteristicprofile according to their electrophoretic mobility when the nucleic acid is run in gel. The aim of this work was to determineif electrophoretic mobility is useful for discovering the genomic diversity of the virus by means of analysis of molecular variance(AMOVA). For 13 crop years between 1990 and 2004, 209 samples of diseased plants with MRCV were collected in varioushosts (maize, oats, wheat, barley, triticale, rye and tricepiro), in 11 locations of ecologically different zones, and submitted toelectrophoretic analysis. A binary matrix was made up where one means the presence of an especific segment at a particularelectrophoretic distance and zero the absence in that position, and AMOVA was applied. Results suggest genomic variabilitybetween and within locations, between and within crop years, but not between the different hosts. This would imply that thebroad and continually growing range of hosts does not influence the diversity of the virus electrophoretic profile. It is hoped thatthis will provide information applicable to population and epidemiological studies of this important disease.
