Resumen:
Angiogenesis, the growth OF new capillariesfrom preexisting blood vessels, is a complexprocess involving endothelial cell (EC) activation, disruptionof vascular basement membranes, and migrationand proliferation of ECs. Glycan-mediated recognitionhas been proposed to play an instrumental rolein mediating cell-cell and cell-matrix interactions. Galectins(Gal), a family of glycan-binding proteins withaffinity for -galactosides and a conserved sequencemotif, can decipher glycan-containing information andmediate cell-cell communication. Galectin-8 (Gal-8), amember of this family, is a bivalent “tandem-repeat”-type galectin, which possesses 2 CRDs connected by alinker peptide. Here, we show that Gal-8 is endowedwith proangiogeneic properties. Functional assays revealeda critical role for this lectin in the regulation ofcapillary-tube formation and EC migration. Moreover,Matrigel, either supplemented with Gal-8 or vascularendothelial growth factor (VEGF), injected in miceresulted in induction of in vivo angiogenesis. Remarkably,Gal-8 was expressed both in the cytoplasm andnucleus in ECs of normal and tumor vessels. Furthermore,CD166 [activated leukocyte cell adhesion molecule(ALCAM)] was identified as a specific Gal-8-bindingpartner in normal vascular ECs. Collectively, thesedata provide the first evidence demonstrating an essentialrole for Gal-8 in the regulation of angiogenesis withcritical implications in tumor biology
