Background: There are no phenotypic methods available to unequivocallydifferentiate C. glabrata complex especies.Objectives: To develop a new multiplex PCR method designed todifferentiate the three species of the C. glabrata complex and to use itto study a C. glabrata collection to uncover strains of the newlydescribed species.Material and methods: The method was developed based on ITS sequencedifferences between the species. It was validated by using a blindedcollection of strains and finally, the new molecular method was used tostudy a collection of 192 C. glabrata complex strains. The obtainedresults were compared with ITS sequencing.Results: The proposed method showed 100% concordance with ITS sequencingand proved to be effective for clinical and epidemiological applications.We found two C. bracarensis and three C. nivariensis out of the 192studied strains (0.93% and 1.40% prevalence, respectively).Conclusions: The presented a fast, inexpensive, robust and highlyreproducible multiplex PCR method showed 100% concordance with ITSsequencing. It usefulness was demonstrated by studying a large collectionof C. glabrata sensu lato strains.
