Papaya ringspot virus W infecting Luffa aegyptiaca in Cuba

Luffa aegyptiaca (sponge gourd) plants showingsevere leaf mosaic and deformation symptoms were observedin Villa Clara, Cuba. Electron microscopy observations of leafdip preparations revealed flexuous filamentous particles,which were identified as Papaya ringspot virus (PRSV) byELISA test. PRSV was mechanically transmitted to healthyCucurbita moschata plants. Sequence analysis of the RT-PCRproduct obtained from the capsid protein gene showed highestidentity with other PRSV isolates from United States ofAmerica, Australia, Venezuela, Brazil, India and Cuba, rangingbetween 93.2?96.5% and 93.4?98.1% for nucleotide andamino acid sequences, respectively. Phylogenetic analysisshowed two main clusters. Cluster I included isolates fromthe Americas-Australia group and India, including spongegourd isolate, while the cluster II included isolates fromChina, Thailand and Taiwan. To our knowledge, this is thefirst report of PRSV-W infecting sponge gourd in Cuba.
