One is an intrinsic aactivity of the phosphoenolpyruvate carboxykinase (PEPCK), shown by the purified enzyme. The other OAD activity corresponds to a protein which can be separates from PEPCK by ion exchange chromatography on Mono Q or by affinity chromatography on ATP-agarose, and shows distinct properties in terms of divalent cation activation and pH optima. Extraction from cells broken by freezing and thawing affords enzyme preparations rich in Cd2+-activated OAD activity and poor in PEPCK, which can be used as a source for the purification of the former enzyme.
