Citrus psorosisvirus (CPsV) and Mirafiori lettucebig-veinvirus (MiLBVV),membersofthe Ophioviridae family,havesegmentednegative-sensesingle-strandedRNAgenomes.Todatenoreportshavedescribed how ophiovirusesspreadwithinhostplantsand/ortheproteinsinvolvedinthisprocess.Hereweshow that the54KproteinofCPsVisencodedbyRNA2anddescribeitssubcellulardistribution.Upontransient expressionin Nicotiana benthamiana epidermal cellsthe54Kprotein,andalsoits54Kcounterpart proteinofMiLBVV,localizetoplasmodesmataandenhanceGFPcell-to-celldiffusionbetweencells.Both proteins,butnotthecoatproteins(CP)oftherespectiveviruses,functionally trans-complement cell-to- cell movement-defective PotatovirusX (PVX) and Tobaccomosaicvirus (TMV) mutants.The54Kand54K proteinsinteractwiththevirus-specific CPinthecytoplasm,suggestingapotentialroleofCPin ophiovirus movement.Thisisthe first studycharacterizingthemovementproteins(MP)ofophioviruses
