Rheological, Physical and Spectroscopical Characterization of Gamma Irradiated Albumin Nanoparticles Loaded with Anthocyanin

Anthocyaninsare the main active compounds in blueberry. However, they have poor oxidationstability. If anthocyanins are encapsulated in protein nanoparticles, theiroxidation resistance could be increased due to the slowing down of theoxidation process. This work describes the advantages of using a γ-irradiatedbovine serum albumin nanoparticle bound to anthocyanins. The interaction wascharacterized biophysically, mainly by rheology. By computational calculationand simulation based on model nanoparticles, we estimated the number ofmolecules forming the albumin nanoparticles, which allowed us to infer a ratioof anthocyanin/nanoparticles. Measurements by UV-VIS spectroscopy, FT-IRspectroscopy, fluorescence spectroscopy, dynamic light scattering (DLS), ζpotential, electron transmission microscopy and rheology at room (25 °C), andphysiological (37 °C) temperatures were performed. The spectroscopymeasurements allowed identifying additional hydrophobic sites created duringthe irradiation process of the nanoparticle. Based on the rheological studies,it was observed that the BSA-NP trend is a Newtonian flow behavior type for allthe temperatures selected, and there is a direct correlation between dynamicviscosity and temperature values. Furthermore, when anthocyanins are added, thesystem increases its resistance to the flow reflected in the morphologicalchanges observed by TEM, thus confirming the relationship between viscosityvalues and aggregate formation
