In the present work, the biosiliceous microremains recovered at the Fuerte SanctiSpiritus Site, the first Spanish settlement to be established in the Río de la Plata basin,and which marks the beginning of European colonization and conquest in America areanalysed. It is located in Puerto Gaboto, Santa Fe province, Argentina and consists ofa brief occupation that took place between May 1527 and September 1529. Theexcavation was carried out according to the open area methodology, for which thesampling was distributed in different features located in the plant of the site. Twentysamples were analysed according to two variables, type of trait and chronology, thatallowed to establish, in accordance with ethnohistorical sources, that the cultivation ofcorn was practised and, in turn, wheat was introduced and probably fattened it.Likewise, the analysis allowed estimating the content of the traits according to the typeof sediment sampled, being able to characterize discard areas, fillings, levels of use,and sediment after the abandonment of the settlement.
