Lactoferrin purification from bovine whey and colostrum by affinity membrane chromatography

Resumen:
Abstract. The utilisation of hollow-fibre membranes derivatised with the dye Red HE-3B for lactoferrin purification from bovine colostrum and whey is studied. Nowadays, membrane chromatography is an interesting industrial alternative where biomolecules are convectively transported towards the ligands immobilised on the pore walls, thus minimising the mass transfer resistance. As a consequence, resolution becomes independent of the flow rate and separations can be performed very quickly at lower costs than that required for chromatography on soft gels. In addition, an extensive clarification of the feed is not required. In this work, the chemistry for attaching Red HE-3B by radiation induced graft polymerisation of acrylic monomers on polysulfone microfiltration hollow-fibre membranes, the opening of the epoxy groups with ammonia and the immobilisation of the dye to the generated amino groups was developed. Moreover, the chromatographic behaviour of the affinity membranes for lactoferrin purification from bovine colostrum and whey was characterised at the equilibrium through the adsorption isotherms and under dynamic conditions after the assembling of a chromatographic cartridge with the modified hollow-fibres. Under dynamic conditions, high purity lactoferrin was obtained with productivity 10 times higher than that achieved with a column of Sepharose with Red HE-3B immobilised. purification from bovine colostrum and whey is studied.
