Analysis of the repetitive sequences in the component genomes of peanut using BAC end-sequences and in-situ hybridization. Guimarães, Patrícia ; Bertioli, Soraya Leal; Seijo, Guillermo ; Town C ; Bertioli, D. J.   Cultivated peanut, Arachis hypogaea is an allotetraploid with an AABB genome. It is of recent and monophyletic origin. A. duranensis and A. ipaënsis are the most probable direct ancestors of cultivated peanut. Advances in the knowledge of the relationships of wild and cultivated genomes are likely to improve our ability to efficiently introgress wild genes into the peanut crop using molecular breeding. Recently we constructed two BAC libraries, one for each of the diploid species. The libraries are respectively c. 7.4 and c. 5.3 genome equivalents with low organelle contamination and average insert sizes of 110 and 100 kb respectively. In order to begin to understand the component genomes of the allotetraploid cultivated peanut we used fluorescent in-situ hybridization (FISH) and analysis of BAC end sequences (BES) from the A. duranensis and A. ipaënsis BAC libraries.
