Resumen:
The combination of Amoxicillin-AMOX and Sulbactam pivoxil-SULB is administrated as tablets and oral suspensions in the treatment of bacterial infections. The quality control of these formulations included, among others, uniformity of content and dissolution test. There is not pharmacopoeial methods to determine Sulbactam pivoxil alone or combined. Although Qi et al.(2003) proposed the simultaneous determination of both drugs using HPLC and UV-determination, the area ratio is very high (>8) what make it difficult to use it mainly in the dissolution test. The purpose of this work is to optimize the operative conditions of a HPLC method for the simultaneous determination of AMOX and SULB, in term of area ratio and retention time.Materials and methods:A HPLC Perkin Elmer-200 with UV detector and a C18 column (150x4.6mm, 5μm) were used. A acetonitrile:water mobile phase was delivered at a flow rate of 1.0 mL.min-1. A 22 design with central point was selected for subsequent optimization. Acetonitrile content in the mobile phase (20-80%) and wavelength of detection (205-230nm) were modified. The retention time, area ratio and heights ratio were analyzed.
