Atriplex undulata is an endemic species from Argentine Patagonia.This study aims to evaluate the broad spectrum of antioxidant and antimutagenic activities of Atriplex undulata (Moq) D. Dietr. (Chenopodiaceae) extracts.Ethanolic extract, different subextracts and isolated compounds showed significant antioxidant activity by DPPH free radical scavenging and crocin bleaching assays.Free and glycosidated flavonoids have been isolated from EtOAc subextract, such as kaempferol, quercetin, rutin, isorhamnetin and isorhamnetin di-glucopyranoside.The protective effect of the extract has been assumed to be due to the strong scavenging and antioxidative properties of those abundant flavonoids and their glycosides.Results indicate that all the flavonoids possess excellent DPPH free radical scavenging activity (IC50 6.07 - 10.79 μg/ml) while crocin bleaching assay showed a K rel ranging from 1.58 to 2.13 μg/ml.These compounds were also tested against mutagenesis using the Amest test with Salmonella typhimurium TA102, which readily responds to reactive oxygen species, and the antimutagenic activity was evaluated against oxidative mutagens terbutyl hydroperoxide and hydrogen peroxide. Trolox was used as a standard in all the experiments.This extract showed antimutagenic activity against t-BuOOH and H2O2 with percent inhibition of mutagenicity ranging from 72.01 % to 96.18 % in a concentration dependent manner. From these results, we conclude that in the Salmonella assay with strain TA102 antimutagenic activities of flavonoids against the peroxide mutagens t BuOOH and H2O2 are mainly caused by radical scavenging effects.Phytochemical analysis by HPLC, LC?MS and total phenolic content revealed high content of isorhamnetin which might be responsible for promising antioxidant and antimutagenic activities of Atriplex undulata extract. Further, contribution of bioactive compounds detected in this study is to be explored to understand the exact mechanism of action as well as their therapeutic efficacy.
