Selection of peptides with affinity for cutinase using a phage display library

A constrained nonapeptide phage display library was selected against the industrially important lipolytic enzyme cutinase in order to find new affinity ligands to this enzyme. After seven cycles of biopanning, 500 clones were isolated and individually tested for their capability to interact with cutinase. The sixteen most promising phage peptide clones were isolated and the genomes of six of them were sequenced. The three phage peptide clones that presented the strongest interaction with cutinase showed the same insert sequence (CRLHHWRYC). Different approaches were made in order to find a practical application for the selected phage clones.
