Although mammalian spermatozoa (Sp) undergo morphological changes during epididymal transit, these have not been studied in mice. The set up of Sp head (h) regions, cholesterol distribution and Sph dimensions were analized. Initial (caput = K) and distal (cauda = C) epididymis were isolated, cut under HM media (Visconti, 1995) at 36ºC and incubated until Sp were released. Suspesions were divided in 2 aliquots, centrifuged (750 g, 10 min) and fixed with PBS (0.01M) and 8% glutaraldehyde (control) or same fixative with fillipin (0.01%) – structural markers of cholesterol – (experimental 1). Fixed Sp were placed onto mica grids, dehydrated and surfaces replica were made. Samples were observed in EM and wide (W), length (L) and head bend (B) were quantified. Sph zones are not defined in K Sp but Sph from C present well defined regions. W:K: X = 3.5µ (±0.2) y C: X = 3.9µ (±0.2); L:K: X= 7µ (±0.6) y C: X = 7.9µ (±0.4) and B:K:X = 4.7µ (±0.6) y C: X = 4.2µ (±0.3). No differences in cholesterol were found. Sh are redimensioned during epididymal transit and the principal sperm head zones, involved in acrosomal reaction and binding with pellucid zone, are there delineated.
