Resumen:
The use of progesterone receptor (PR) ligands for breast cancer treatment remains controversial. We propose that antiprogestins inhibit the growth of tumors with higher levels of isoform A (PRA) than isoform B (PRB) and progestins may inhibit those with opposite ratios. We developed in vivo metastatic models to evaluate PR ligands effects focusing on NDRG1, a progesterone (Pg)/MYC regulated gene. We used a luminal murine mammary carcinoma (C7-2-HI), triple negative human breast cancer MDA-MB-231 cells transfected with PRB (MDA-MB-231-PRB), and the inducible MDA-MB-231-iPRAB cells, which express PRB or PRA under doxycycline or RSL1 administration. When C7-2-HI tumors reached 25 mm2, pellets containing progesterone (Pg; 10 mg every 10 days), medroxyprogesterone acetate (MPA; 20 mg), telapristone acetate (TLP; 6 mg) or mifepristone (MFP; 6 mg) were implanted.
