Shiga toxin-producing Escherichia coli in raw ground beef and environmental samples from retail stores form Argentina: improvement actions

In Argentina postenteric hemolytic uremic syndrome is endemic and the most frequent cause is Shiga toxin-producing Escherichia coli (STEC) O157:H7, O145:[H27,H-,NT], O121:H19, O26:[H2,11,NT], O111:[H-,NT] and O103:[H2,H-,NT] serogroups. The aim of this work was to reduce the presence of STEC in retail stores. The study was divided in three stages: a) estimation of STEC presence in retail stores; b) implementation of improvement actions to reduce STEC; and c) verification of program success. During 2011-2013 period, 86 retail stores from Argentina were evaluated, including 172 raw ground beef (RGB) and 672 environmental samples (E). All samples were processed for STEC O157 isolation and for non-O157 detection by real time PCR and isolation. During 2011, 11 and 7 STEC O157:H7 strains were isolated from RGB and E samples, respectively. Eleven and 28 non-O157 STEC strains were isolated from RGB and E samples,respectively. In 2012, a good manufacturing practices (GMP) program was implemented in all the retail stores. During 2013, 3 and 2 STEC O157:H7 strains were isolated from RGB and E samples, respectively. Eleven and 22 non-O157 STEC strains were isolated from RGB and E samples, respectively. All STEC O157:H7 strains isolated in 2011-2013 period were stx2+stx2c(vha)/ exhA/eae/efa/toxB. The most prevalent non-O157 STEC serotypes in order offrequency were: O174:[H21,28], O178:H19, O8:H19, O91:H21, O171:H14, O113:H21, O116:H21, O41:H49, O79:H19. The most frequent genotype was stx2/saa/exhA/iha. The improvement actions implemented were successful due to the reduction of STEC O157:H7. Although one O26:H11 STEC strain was isolated in the verification stage, the non-O157 STEC strains isolated are intimin-negative, and do not belong to the serogroups associated to human disease in Argentina. It is important to highlight that the implementation of systematic monitoring, work training and awareness of food handlers made not only improve product quality butalso the environment where food is handled.
