Low temperature is one of the most important factors affecting plant growth, directly altering the photosynthetic process andleading to photo-inhibition.With the aim to address thephotosynthetic acclimation response of L. japonicus under coldstress, two ecotypes (MG-1and MG-20) were studied. Previousdata showed that photo-inhibitionoccurs in stress anddifferentially between ecotypes, being MG-1more affected thanMG-20.In this study, we used a proteomic approach to evaluatechanges in the chloroplast stroma of both ecotypes after 24 h oflow temperature. A total of 450 proteins were identified, with 325uncharacterized in the L. japonicus protein database.Proteinswere classified and 168 and 86 were differentially accumulatedin MG-1and MG-20, respectively, in the stress treatmentcompared to controls. Of these, only 3 and 4 proteins were moreabundant, while 165 and 82 showed a reduction in itsaccumulation at low temperature. A functional annotation andclassification was made by Gene Onthology, KEGG MetabolicPathways and blastp. The results obtained showed that down-regulated proteins were mainly related to amino acid, carbohydrate and energy metabolisms. However, particular metabolic pathways were different regulated in each case and for each ecotype. Data obtained may be useful in identifying proteins with a key role in low temperature acclimation in legumes.
