They hydrolyze the heterosidic linkage of diglycoconjugates such as rutin or hesperidin, generating a disaccharide and the corresponding aglycon as products. Some diglycosidases exhibit transglycosylation activities, which provide access to various novel diglycoconjugates (Figure 1). Figure 1. Hydrolysis and transglycosylation reactions of some microbial diglycosidases We report on the cloning of several diglycosidase-encoding genes from three fungal strains: Mucor circinelloides, Penicillium chrysogenum and Acremonium sp. We succeeded in heterologously expressing some of these diglycosidases in active form using Escherichia coli or Pichia pastoris as a host. This enabled us to characterize these enzymes in terms of their substrate specificities and transglycosylation activities.
