Resumen:
In hypertension there is an exacerbation of the classic pressor arm of the renin angiotensin system (RAS) in detriment of the depressor arm constituted by angiotensin converting enzyme 2/angiotensin (Ang)(1-7)/Mas receptor (R). Our aim was to evaluate if a differential regulation of the neuronal Mas R contributes to an unbalanced RAS and therefore to the development of hypertension in spontaneously hypertensive rats (SHR). Methods: Primary culture of neurons (N) from hypothalamus and brain stem of SHR and normotensive rats (Wistar Kyoto-WKY) were incubated in the presence or absence of Ang(1-7) (10-6M) during 15 or 30 min. MasR expression was evaluated by western blot (WB) and immunofluorescence in non-stimulated N, and MasR trafficking by colocalization with Lysotracker, a lysosome marker, or Hoescht, a nuclear marker.
