EFFECT OF BIOMIMETIC MATRICES ON OSTEOGENIC DIFFERENTIATION OF HUMAN MESENCHYMAL STEM CELLS

Mesenchymal stem cells (MSCs) accelerate osseointegration of bone grafts and improve efficiency and uniformity in the formation of bone tissue. MSCs are isolated from easily available and abundant sources, providing a practical and clinically attractive approach in bone tissue regeneration. Our aim was to study the effect of nanofibrous biomimetic matrices composed of poly-ε-caprolactone (PCL) and nanohydroxyapatite (nHA) on MSCs adipogenic differentiation. MSCs isolated from adipose tissue plastic surgeries samples were deposited above biomimetic matrices of PCL/nHA in 10% FBS supplemented DMEM. We evaluated adhesion, penetration and differentiation of these cells cultured over nanofibrous matrices. To induce osteoblast differentiation, we supplemented the culture medium with a cocktail containing 5% horse serum, 10 mM β-glycerophosphate, 0,1 µM dexamethasone and 50 µM 2-phospho-L-ascorbic acid. Osteogenic differentiation was assessed by measuring alkaline phosphatase (ALP) activity, which increased over time and its relative activity was higher in drug cocktail treated cells. MSCs were capable to penetrate the PCL/nHA scaffolds as observed in cross sections analyzed by transmission electron microscopy. These results are our first steps on the transition from monolayer to 3D-nanofibrous biomimetic scaffolds cultures.
