Resumen:
CAPACITY OF MAYORITARY COMPOUNDS FROM ACHYROCLINE SATUREIOIDES TO PROTECT THE DAMAGE INDUCED AFLATOXIN B1 IN RATS WISTARSabini MC., Escobar FM., Cariddi LN., Bagnis G., Comini L., Menis Candela F., Magnoli A., Barberis C., Sabini LI., Dalcero A.Aflatoxin B1 (AFB1) is produced by A. flavus and A. parasiticus. This mycotoxin is carcinogenic, teratogenic, hepatotoxic, immunotoxic. It is the most important mycotoxin worldwide for its effect on public health and animal productivity mycotoxin. Macela has many scientifically proven medicinal properties, such as antioxidant, antimicrobial, others. Main compounds of this species are luteolin, quercetin and chlorogenic acid. The objective was to evaluate the ability of majority compounds (MC) of A. satureioides to protect of genotoxic and liver damage induced by AFB1 in rats.Wistar rats of 200 g were used; lots of 4 individuals (2 males and 2 females) were formed. Animals were inoculated by intraperitoneal injection with a single dose (0.5 ml). The treatments were: AFB1 1- (1 mg / kg body weight) plus L (2.5 mg / kg p.c.); AFB1 2- (1 mg / kg p.c.) more Q (2.5 mg / kg p.c.); AFB1 3- (1 mg / kg) more CLA (5 mg / kg p.c.); Negative control: saline solution; 4- Positive control: cyclophosphamide 30 mg / kg p.c .; 5- AFB1 Control: AFB1 (1 mg / kg p.c.) dissolved in methanol 5% saline. Animals were sacrificed by cervical dislocation at 24h post-injection. It was performed: 1-Micronuclei in mouse bone marrow: by Schmidt W. (1975) number of micronuclei in 1000 polychromatic erythrocytes (EPC) and toxicity index (IT) considering polychromatic erythrocytes (EPC) / normochromatic (ENC) EPC 1000 were determined. 2- Hystopathology: Parts of the organs were pre-served in 10% buffered formaldehyde (pH 7.4). These samples were cut at 4 µm thickness and subjected to haematoxylin/eosin (H&E) staining for microscopic histological examination under 400 × magnifications. Photomicrographs were taken with a Zeiss Axiostar plus microscope and analyzed. The results of genotoxicity revealed that AFB1 at 1 mg/kg showed an index of genotoxicity of 35 MN/1000 EPC, showing statistically significant difference with the negative control (p
