Risk-assessment of Nicotiana tabacum plant extracts expressing two different recombinant heterologous proteins in female Balb/c mice

The genetic (stable) and transient expression in plants is a useful tool to produce recombinant proteins which can be delivered in foliar extracts. The use of these novel delivery systems need deep risks-evaluations demonstrating the non-toxicity of the recombinant foliar extracts per se in order to confirm that this methodology is not harmful for animals. The present work takes into account the assessment of the infected plant and the foliar extract injected into mice to deliver plant made heterologous proteins. Mice Balb/c were subcutaneously immunized by administering foliar extracts from genetic modified plants expressing the full antibody r14D9 or the glycoprotein tE2 from VBDV transiently expressed, both formulated with complete Freund?s adjuvant. Three boosters of each foliar extract were applied. Al(OH)3- Hydrogel® was used as an alternative adjuvant. Control animals were primed and boosted with non recombinant tobacco foliar extracts plus each one of both adjuvant. No significant differences between treatments were observed. The classic pathologic lessons by inflammatory processes triggered by adjuvants were obtained in all mice injected. The histological sections of the inoculation sites, as well as the main organs, showed no additional lesions. Clinical and haematological results are in coincidence with the support the observations previously described.
