A variety of cellular processes taking place at first stages of pregnancy are encompassed to ensure proper placentation in a predominant anti-inflammatory and immunosuppressant microenvironment. Trophoblast cells orchestrate leukocyte recruitment and functional shaping to maintain immune homeostasis. We have previously shown that vasoactive intestinal peptide (VIP) favors a suitable anti-inflammatory and immunosuppressant profile for placentation and fetal growth in different murine models and human cell settings. Here we studied pregnancy outcome in VIP KO mice, particularly the relevance of endogenous VIP at the early maternal-placental interface and its impact on the local expression of immune and trophoblast functional markers. Implantation sites isolated from C57BL/6 WT mated to VIP-/- or WT males at day 8,5 of pregnancy were analyzed by RTqPCR, immunofluorescence or laser capture microdissection (LCM). Trophoblast giant cells (TGC) isolated by LCM were analyzed for VPAC1 and VPAC2 expression by RTqPCR in parallel to MMP9 and Prl3d1 TGC markers.Implantation site explants from WT mothers crossed with VIP -/- males presented structural differences and decreased IL-10 and Foxp3 expression compared with WTxWT matings (P
