Differential effects of lactic acid bacteria peptidoglycans on lung epithelial cells and alveolar macrophages

We demonstrated that peptidoglycan from Lactobacillus rhamnosusCRL1505 (Pg05) is as effective as viable strain for improvingsystemic and respiratory humoral immune response against Streptococcuspneumoniae in malnourished mice. These immunomodulatorycapacity cannot be extrapolated to peptidoglycans (Pgs)from other strains. The aim of this work was to study the interactionsof Pgs obtained from different lactobacilli, with mice lung epithelialcells (LEC) or alveolar macrophages (AM), in order to elucidatethe mechanisms involved in the differential effect of Pg05. We comparedPg05 with Pgs from the non-probiotic L. rhamnosus CRL534(Pg534) and from the probiotic L. plantarum CRL1506 (Pg06). AMor LEC were cultured 24h or 24-48h with different concentrations ofPg05, Pg06 or Pg534, in the absence or presence of S. pneumoniae.In addition, we studied the cytokines production of LEC withconditioned medium from AM pre-stimulated with Pgs. The resultsshowed that 90% of LEC and AM remained viable after the treatmentswith Pgs. Pg05 (50 ug/mL) and Pg06 were able to differentiallymodulate the cytokines production in AM and LEC.
