IMMUNOMODULATORY PROPERTIES OF PEPTIDOGLYCANS OBTAINED FROM DIFFERENT Lactobacillus STRAINS IN IMMUNOCOMPROMISED-MALNOURISHED MICE

Background: Lactobacillus rhamnosus CRL1505 peptidoglycan (Pg05) improves innate immune response in immunocompromised-malnourished mice. The aim of this work was to evaluate whether the immunoregulatory activity of Pg05 is a strain specific property. Then, the effects of three peptidoglycans on the immune response to pneumococcal infection were examined. In addition to Pg05, peptidoglycans from probiotic L. plantarum CRL1506 (Pg06) and the non-immunomodulatory control L. rhamnosus CRL534 (Pg534) were evaluated.Methods: Weaned mice were malnourished with a protein-free diet for 21 days and then received balanced diet during 7d (BD group) or BD with nasal supplementation of Pg05, Pg06 or Pg534 during the last 2d. On d8, all groups were infected with Streptococcus pneumoniae (Sp) and the resistance to the infection and innate immune response were studied.Results: Lung and blood Sp counts in Pg06 and Pg534 groups were similar to BD group while the pathogen was not detected in samples of Pg05-treated mice. Moreover, lung tissue injuries in Pg05 were significantly lower when compared to the other groups. Pneumococcal infection increased CD45+Gr-1+ neutrophils, CD45+F4/80+CD11b+CD11c- macrophages, TNF-a and IL-10 in the respiratory tract of all groups. However, the increment of phagocytes numbers as well as their MHC-II expression were higher in Pg05 mice than the other groups. In addition, Pg05 mice showed a lower TNF-a/IL-10 ratio when compared to Pg06 and Pg534 groups.Conclusions: The immunomodulatory effect of Pg05 is a strain specific property and, this particular feature of Pg05 opens interesting questions regarding the molecular differences between probiotic peptidoglycans.
